La version 5 du Test de Suspension Caudale de Bioseb, basée sur des capteurs de force ainsi que sur de l'acquisition vidéo, est la seule et unique solution totalement automatisée au monde pour effectuer un test de désespoir comportemental simple et objectif sur souris. Il s'agit d'une méthode rapide, fiable et sans douleur pour tester les effets psychotropes de drogues et substances comme les antidépresseurs et les sédatifs. Ce test classique et indolore pour la recherche sur l'anxiété et la dépression est désormais présenté avec un nouveau logiciel puissant et moderne, dans une version sans-fil BlueTooth.

Présentation
Le test de suspension caudale a été développé en tant qu'alternative au test de nage forcée de Porsolt, et repose sur un concept similaire. Des souris, suspendue par la queue en utilisant un morceau de ruban adhésif (une méthode sans douleur), essaient, de manière innée, de s'échapper de cette situation non-naturelle. A force d'échouer dans leur tentative d'échappement, elles génèrent un état de désespoir qui se traduit par de l'immobilité. La magnitude de cette immobilité est considérée comme corrélée à un état dépressif, et est réduit de façon significative par les antidépresseurs. Le Test de Suspension Caudale a été décrit dans les années 80 par Steru et al. et permet l'évaluation rapide des effets psychotropes de drogues et substances (antidépresseurs, sédatifs), ce qui en fait un test classique et indolore pour la recherche sur l'anxiété et la dépression.
Le TST 4 de Bioseb est le seule test complètement automatique au monde! Il est basé sur le dessin original, le 'ITEMATIC-TST' décrit par Steru en 1987, et utilise à la fois des capteurs de force et un enregistrement vidéo. Le test ne génère aucune douleur sur l'animal: la queue de la souris est simplement fixée au capteur en utilisant un morceau de ruban adhésif.
Le dessin original ITEMATIC a été modifié, adapté et amélioré pour la compagnie Porsolt and Partners Pharmacology en 2000. L'instrument peut accommoder 3 souris dans 3 compartiments différents, et propose une hauteur au sol réglable, ainsi que des plaques anti-escalade pour les souris de lignée C57.
Le test de suspension caudale de Bioseb est un instrument offrant des fonctionnalités uniques, telles que la randomisation de la liste d'animaux, la mesure de 6 animaux simultanément, et le calcul direct des paramètres d'immobilité, d'énergie et de puissance en mouvement. L'utilisateur peut modifier les réglages pour s'adapter à son protocole de recherche. La valeur d'énergie est une méthode unique permettant de différentier le balancement passif et la lutte active. La puissance en mouvement est un calcul discriminant supplémentaire donnant une indication de la force de la souris.
Comme ça marche?

Le principe de mesure est basé sur la quantité de mouvement générée par une souris essayant d'échapper à sa suspension. Durant le test de suspension caudale, les mouvements des souris sont analysés en terme d'activité (active/non-active), d'énergie et de puissance développée dans le temps. Dans la mesure où l'observation humaine est fastidieuse, peu répétable, et prend du temps, le test de suspension caudale de Bioseb (TST) automatise l'intégralité de l'expérience.
L'opérateur prépare ses morceaux de ruban adhésif pour suspendre les souris et les cages contenant les animaux à tester. Une fois que l'expérience a été configurée via le logiciel et que la liste des animaux a été randomisée, l'opérateur suspend chaque souris individuellement en suivant la randomisation affichée à l'écran. Chaque voie est ensuite démarrée de manière individuelle en pressant une touche dédiée, afin de s'assurer que chaque animal est suspendu pour la même durée (ce qui est impossible avec un système basé uniquement sur la vidéo).
Lorsque le cycle expérimental est terminé, les souris sont retirés et le système demande à l'opérateur de suspendre le prochain groupe d'animaux. Après l'expérience, la totalité des groupes peut-être analysée, et des statistiques sur les minimums et maximums, moyennes, ainsi que sur les déviations standards, sont effectués en quelques secondes. Les données sont présentées de manière globale par groupe et détaillées pour chaque groupe par individus (souris).
Le logiciel TST-SOFT génère également des graphiques qui peuvent être exportées au format BMP pour animer de futures présentations scientifiques.
