En tant que version électronique du classique esthésiomètre à filaments de Von Frey, la dernière évolution de l’instrument électronique Von Frey de Bioseb, conçu pour déterminer le seuil de sensibilité mécanique chez les rongeurs (rats et souris) par stimulation sous la patte, est un outil indispensable pour vos recherches sur l’hyperalgésie et l’allodynie. En mesurant et en enregistrant la force à laquelle l’animal présente un réflexe de retrait de la patte, il est possible d’étudier les pathologies liées à la réponse sensorielle ainsi qu’à l’hyper- ou l’hypoesthésie.

Présentation

16 ans après la création du premier Von Frey Electronique de Bioseb, nous révolutionnons l’utilisation de ce test standard pour la mesure du seuil de sensibilité à la douleur mécanique chez les rongeurs (rats et souris).
Bien connu pour ses difficultés d’utilisation lorsque l’on veut stimuler sous la patte des rongeurs, le test de Von Frey Electronique est généralement critiqué par les opérateurs pour les troubles liés au fait de devoir se pencher et pour la position inconfortable à maintenir pour effectuer son geste. Malgré l’utilisation de postes de travails adaptés (sièges bas, stands surélevés...), la manipulation reste difficile, voire douloureuse lorsque répétée sur un grand nombre d’animaux.
Afin d’y remédier, nous avons intégré une caméra au niveau de la poignée d’utilisation de notre appareil pour pouvoir filmer directement par-dessous les pattes. Couplé à l’utilisation d’un logiciel dédié, nous avons repensé la manière dont l’opérateur peut se positionner pour effectuer le test.
Utilisé sur une tablette tactile, le logiciel EVF 5 permet ainsi de visionner le flux vidéo en temps réel sans que l’opérateur ait à se pencher. De plus, il intègre l’ensemble des fonctionnalités qui peuvent être recherchées pour ce test :
• La gestion des listes d’animaux.
• L’affichage en temps réel de la courbe de force en fonction du temps pour améliorer son geste et la reproductibilité des mesures.
• Un tableau permettant d’assigner une mesure à la patte correspondante, directement en cliquant sur l’écran tactile.
• Une relecture à la fois des vidéos et de la courbe de force pour chacune des mesures afin d’éliminer celles qui ne conviennent pas.
• Un export des résultats dans un tableau Excel.
Principes de fonctionnement
Composé d’une poignée de stimulation reliée à une tablette tactile intégrant un logiciel dédié, l’appareil est pensé pour que l’opérateur n’ait pas à se pencher et puisse avoir un environnement complet pour le Test de Von Frey.
L’opérateur crée une expérience sur le logiciel dédié et paramètre sa liste d’animaux ainsi que les différentes pattes qu’il souhaite stimuler (généralement pattes arrières). Il peut ensuite démarrer le test en prenant en main la poignée de stimulation et observe sous les pattes des rongeurs directement à travers le flux vidéo sur la tablette tactile et non en se penchant sous les pattes.
La stimulation est généralement effectuée sur les pattes arrières du rongeur. Le retrait de la patte causé par la stimulation mécanique est enregistré comme la réponse à ce stimulus. Lors de la mesure, l’opérateur peut suivre en direct et ainsi adapter la force appliquée en fonction du temps.
La force maximale appliquée avec l’appareil, correspondant à la valeur nécessaire pour le retrait de la patte, est sauvegardée par le système. L’opérateur peut directement décider de valider cette mesure en cliquant sur l’écran tactile (voir photo dans la partie logiciel ci-dessous) ou relancer la mesure avec une nouvelle stimulation.
