Comparison of Ginseng Leaf Extract and Its Acid-Treated Form- UG0712 Between Their Effects on Exercise Performance in Mice

Abstract
C2C12 myotube cells were treated with GLE or UG0712 as in the experiment of the cell viability assay above. Subsequently, the cells were collected and were lysed using RIPA buffer (LPS Solution, Dajeon, Korea). The resultant cell lysates were sonicated on ice for 10 seconds 3 times, and debris are removed by centrifugation (12,000 x g) for 20 minutes at 4C. Proteins (25-40 g) in the supernatants were separated in 8%-12% SDS-polyacrylamide gel and transferred to PVDF membranes of 0.45 m pore size. To mitigate nonspecific binding, the membranes were blocked with 5% skim milk solution in 1 hour at room temperature. Then, the membranes were probed with the following primary antibodies specific to AMP-activated protein kinase (AMPK), phosphorylated-AMPK, and nuclear respiratory factor-1 (NRF-1) (all of these antibodies were obtained from Cell Signaling Technology, Danvers, MA, USA), peroxisome proliferator-activated receptor gamma coactivator-1 alpha (PGC-1Alpha), mtTFA, transcription factor A, mitochondrial (TFAM), and cyclophilin B (CypB) (all from Santa Cruz Biotechnology (Dallas, TX, USA) and sirtuin 1 (SIRT-1) (Abcam) for overnight at 4C, after which the membranes were incubated with peroxidase-conjugated secondary antibodies for 1 hour at room temperature. The protein bands were visualized using enhanced chemiluminescence reagents (Bio-Rad Laboratories) on an ImageQuant LAS-4000 imager (General Electric, Trevose, PA, USA) and quantified using the Image J software (NIH, Bethesda, MD, USA).24The band densities of proteins were normalized to that of CypB and the normalized values were compared.
