Assessment of Postoperative Analgesic Efficacy and Animal Well-Being Using a Novel Triaxial Accelerometer Device- the Rodent Fitbit-Like Telemetry Device

Abstract
MSR, a cyclooxygenase-2 (COX2) enzyme inhibitor with some anti-COX1 properties, is a commonly used sustained-release analgesic in laboratory animals. While this sustained-release nonsteroidal antiinflammatory drug is marketed as providing up to 72 h pain relief in several laboratory species, such as dogs, swine, sheep, and macaques,42concerns have emerged regarding whether the previously suggested dose of 6 mg/kg would be sufficient in providing analgesic coverage in mice.23In our study, we demonstrated that mice without surgical manipulation did not display any significant alterations in their activity levels, regardless of sex. In contrast, a previous study43evaluating a higher concentration of meloxicam did identify decreases in exploratory activity. Moreover, we found that MSR mice which had undergone paw incision did not demonstrate any significant change in activity throughout the study. Mechanical hypersensitivity was unsuccessfully attenuated in B6 mice for the entire duration of the study. It has been previously reported43,44that mice provided MSR have an increased frequency of developing erythematous and/or mass-like lesions. While we did appreciate sporadic development of mass-like to ulcerative lesions adjacent to the drug injection site in 5 of our MSR-treated mice after device removal, gross necropsies were not performed on all the animals to confirm and assess these lesions histologically. Interestingly, we did document 2 cases of severe paw edema postoperatively in mice provided only MSR after surgical manipulation that failed to resolve by the end of the study. At the conclusion of the study, one mouse was submitted for gross necropsy and histology to assess the edema noted on the surgical paw. The necropsy report revealed several findings. With respect to the injection site area, the nape of the neck demonstrated severe, chronic, locally extensive necrosuppurative epidermatitis with fibrosis. The ipsilateral tibial musculature had mild to moderate, multifocal, chronic myofiber degeneration and regeneration. The right hindpaw was noted to have severe, locally extensive, neutrophilic dermatitis with myositis, fasciitis, and panniculitis, along with predominantly plantar dermal fibrosis containing granulation tissue formation serocellular crusting, ulceration, and acanthosis with bone remodeling of the proximal phalanx and woven bone formation. While slight edema of the ipsilateral paw was observed in all mice (irrespective of sex and treatment group), it would typically resolve by the second day postsurgery. We have not identified any additional studies documenting this type of adverse clinical presentation in MSR-treated mice used for paw incisional pain studies. The presence of poor wound healing postadministration of an NSAID has been reported45,46to interfere with healing in musculoskeletal injuries through inhibition of muscle repair properties from COX-1 and COX-2 pathways and decrease bone formation. Studies into the effects of MSR on musculoskeletal injuries and healing were outside the scope of this study and merit further investigation.
