Abstract
Studies on the composition of gut microbiota in relation to sarcopenia have been increased over the past few years [48-50]. At the family level,LachnospiraceaeandRuminococcaceaeare known for their roles in gut health and metabolism, with lower levels observed in individuals with sarcopenia [51]. In addition, the familyCoriobacteriaceae, which includes the genusCollinsella, has been noted for its potential pathogenic role and altered abundance in patients [52]. At the genus level,PrevotellaandCoprococcushave been identified as significant markers, with research showing their reduced abundance in sarcopenic populations. In our study, a higher Shannon index was observed in the ATG-F4 supplemented group than in the DEX group, indicating that ATG-F4 supplementation modulates the composition of the gut microbiota toward greater evenness [53]. The PCoA further supported this finding, showing distinct clustering patterns indicative of compositional alterations in the gut microbiome following ATG-F4 supplementation. These changes were characterized by an increase inLachnospiraceaeand a marked decrease inFaecalibaculumandFirmicutes. Notably, ATG-F4 supplementation increased the abundance of beneficial bacteria identified by LEfSe, includingLimosilactobacillus,Bifidobacterium,Lactobacillus,Lacticaseibacillus, andButyricicoccus[54-57]. These bacteria promote gut health and regulate systemic metabolism through the production of short-chain fatty acids and other bioactive compounds. Importantly, alterations in gut microbiota composition following ATG-F4 supplementation were associated with improvements in skeletal muscle phenotypes observed in this study. Specifically, the enrichment of taxa involved in amino acid and lactate metabolism was accompanied by preservation of muscle mass, fiber cross-sectional area, and grip strength, along with suppression of FOXO-dependent proteolytic signaling and restoration of mitochondrial biogenesis–related pathways in skeletal muscle. DEX-induced muscle atrophy was accompanied by reduced microbial diversity and compositional dysbiosis (Fig. 5A-5C), whereas ATG-F4 supplementation partially restored microbial diversity and increased the abundance of bacterial genera involved in amino acid and lactate metabolism, as identified by LEfSe analysis (Fig. 5D).
